@IdkWhyImHere as you can see, at the very first chapter you saw nergal, mephisto,and the other. Nergal ordered mephisto to make Leon drink something. By using his tentacles. How can I tell if it was a drink? Well, based on my observation, Leon said it's a fluid? So I can assume that mephisto made Leon drink something to make Leon pregnant.
